3-Acetoxy-fatty acid isoprenyl esters from androconia of the ithomiine butterfly Ithomia salapia ArchiMer
Mann, Florian; Szczerbowski, Daiane; De Silva, Lisa; Mcclure, Melanie; Elias, Marianne; Schulz, Stefan.
Male ithomiine butterflies (Nymphalidae: Danainae) have hairpencils on the forewings (i.e. androconia) that disseminate semiochemicals during courtship. While most ithomiines are known to contain derivatives of pyrrolizidine alkaloids, dihydropyrrolizines or g-lactones in these androconia, here we report on a new class of fatty acid esters identified in two subspecies, Ithomia salapia aquinia and I. s. derasa. The major components were identified as isoprenyl (3-methyl-3-butenyl) (Z)-3-acetyloxy-11-octadecenoate, isoprenyl (Z)-3-acetyloxy-13-octadecenoate (12) and isoprenyl 3-acetyloxyoctadecanoate (11) by GC/MS- and GC/IR-analyses, microderivatizations, and synthesis of representative compounds. The absolute configuration of 12 was determined to be R. The...

A di-O-dihydrogeranylgeranyl glycerol from Thermococus S 557, a novel ether lipid, and likely intermediate in the biosynthesis of diethers in Archaea ArchiMer
Gonthier, Isabelle; Rager, Marie-noëlle; Metzger, Pierre; Guezennec, Jean; Largeau, Claude.
The lipids of a deep-sea hydrothermal vent arch ae on, Thermococcus S 557, were isolated, purified and structurally determined. Based on acid methanolysis and spectroscopic studies, the polar lipids were shown to comprise diphytanyl glycerol and dibiphytanyl diglycerol, typical membrane lipids of Arch aea. From the neutral lipids, 2,3-di-O-dihydro-14,15-geranylgeranyl glycerol was isolated. This novel ether lipid is, very likely, a close intermediate in the biosynthesis of diphytanyl glycerol diether in Arch aea.

Biochemical survey of the polar head of plant glycosylinositolphosphoceramides unravels broad diversity ArchiMer
Cacas, Jean-luc; Bure, Corinne; Furt, Fabienne; Maalouf, Jean-paul; Badoc, Alain; Cluzet, Stéphanie; Schmitter, Jean-marie; Antajan, Elvire; Mongrand, Sébastien.
Although Glycosyl-Inositol-Phospho-Ceramides (GIPCs) are the main sphingolipids of plant tissues, they remain poorly characterized in term of structures. This lack of information, notably with regard to polar heads, currently hampers the understanding of GIPC functions in biological systems. This situation prompted us to undertake a large scale-analysis of plant GIPCs: 23 plant species chosen in various phylogenetic groups were surveyed for their total GIPC content. GIPCs were extracted and their polar heads were characterized by negative ion MALDI and ESI mass spectrometry. Our data shed light on an unexpected broad diversity of GIPC distributions within Plantae, and the occurrence of yet-unreported GIPC structures in green and red algae. In monocots,...

Bufotenine is able to block rabies virus infection in BHK-21 cells J. Venom. Anim. Toxins incl. Trop. Dis.
Vigerelli,Hugo; Sciani,Juliana Mozer; Jared,Carlos; Antoniazzi,Marta Maria; Caporale,Graciane Maria Medeiros; Silva,Andréa de Cássia Rodrigues da; Pimenta,Daniel C.
Background Rabies is a fatal zoonotic neglected disease that occurs in more than 150 countries, and kills more than 55.000 people every year. It is caused by an enveloped single stranded RNA virus that affects the central nervous system, through an infection initiated by the muscular nicotinic acetylcholine receptor, according to many authors. Alkaloids, such as acetylcholine, are widespread molecules in nature. They are present in numerous biological fluids, including the skin secretion of many amphibians, in which they act (together with proteins, peptides and steroids) as protection agents against predators and/or microorganisms. Among those amphibians that are rich in alkaloids, there is the genus Rhinella. Methods Bufotenine was isolated from Rhinela...

Can the hemoglobin characteristics of vesicomyid clam species influence their distribution in deep-sea sulfide-rich sediments? A case study in the Angola Basin ArchiMer
Decker, Carole; Zorn, N.; Le Bruchec, J.; Caprais, Jean-claude; Potier, N.; Leize-wagner, E.; Lallier, F. H.; Olu, Karine; Andersen, A. C..
Vesicomyids live in endosymbiosis with sulfur-oxidizing bacteria and therefore need hydrogen sulfide to survive. They can nevertheless live in a wide range of sulfide and oxygen levels and depths, which may explain the exceptional diversity of this clam family in deep-sea habitats. In the Gulf of Guinea, nine species of vesicomyid clams are known to live in cold-seep areas with pockmarks from 600 to 3200 m deep, as well as in the organic-rich sediments of the Congo deep-sea fan at 5000 m deep. Our previous study showed that two species living in a giant pockmark have different oxygen carriers, suggesting different adaptations to hypoxia. Here, we studied the hemoglobin structure and oxygen affinity in three other species, Calyptogena valdiviae, Elenaconcha...

Genome sequencing of various organisms allow global analysis of gene expression, providing numerous clues on the biological function and involvement in the biological processes studied. Proteomics is a branch of molecular biology and biotechnology that has undergone considerable development in the post-genomic era. Despite the recent significant advancements in proteomics techniques, still there is much to be improved. Due to peculiarities to the plant kingdom, proteomics approaches require adaptations, so as to improve efficiency and accuracy of results in plants. Data generated by proteomics can substantially contribute to the understanding and monitoring of plant physiological events and development of biotechnological strategies. Especially for...

Characterization of New Oligosaccharides Obtained by An Enzymatic Cleavage of the Exopolysaccharide Produced by the Deep-Sea Bacterium Alteromonas infernus Using its Cell Extract ArchiMer
Akoumany, Katy; Zykwinska, Agata; Sinquin, Corinne; Marchand, Laetitia; Fanuel, Mathieu; Ropartz, David; Rogniaux, Hélène; Pipelier, Muriel; Delbarre Ladrat, Christine; Colliec Jouault, Sylvia.
Bacteria from deep-sea hydrothermal vents constitute an attractive source of bioactive molecules. In particular, exopolysaccharides (EPS) produced by these bacteria become a renewable source of both biocompatible and biodegradable molecules. The low molecular weight (LMW) derivatives of the GY785 EPS produced by the deep-sea hydrothermal vent strain Alteromonas infernus have previously displayed some biological properties, similar to those of glycosaminoglycans (GAG), explored in cancer and tissue engineering. These GAG-mimetic derivatives are obtained through a free radical depolymerization process, which could, however, affect their structural integrity. In a previous study, we have shown that A. infernus produces depolymerizing enzymes active on its own...

Convenient preparation of picolinyl derivatives from fatty acid esters ArchiMer
Dubois, Nolwenn; Barthomeuf, Chantal; Berge, Jean-pascal.
It is essential to have simple rapid methods for the determination of fatty acid structures. Traditionally, fatty acids are analysed by gas chromatography using their methyl ester derivatives (FAME). However, their corresponding mass spectra exhibit molecular ions but are usually devoid of ions indicative of structural features and, notably, the position of double bounds on the aliphatic chains [1]. In the most useful approach to structure determination, the carboxyl group is derivatised with a reagent containing a nitrogen atom. Recently, a convenient method for preparing picolinyl esters from intact lipids has been published [2]. However, some problems occurred in our laboratory when this method was used, leading to some modifications and optimisation....

Criblage bioguidé et analyse CL - SMHR des toxines et d'au tres métabolites d'intérêt chez les dinoflagellés G ambierdiscus et Fukuyoa ArchiMer
Pisapia, Francesco.
The epi-benthic dinoflagellates Gambierdiscus and Fukuyoa produce ciguatoxins (CTXs) and maitotoxins (MTXs), which are among the most potent marine toxins known. CTXs are bio-accumulated and bio-transformed along the marine food chain and cause Ciguatera Fish Poisoning (CFP), the most common non-bacterial foodborne intoxication worldwide. Recently, CFP has been reported from areas previously not considered endemic, namely the Canary Islands, and an increasing number of species has been discovered in these genera. Little is known about the relative toxicity between different species and the toxin congeners produced by these dinoflagellates, especially by strains outside the Pacific Ocean. Several strains of Gambierdiscus and Fukuyoa were screened for their...

Geiger, Marie; Guitton, Yann; Vansteelandt, M.; Kerzaon, I.; Blanchet, Estelle; Du Pont, T. Robiou; Frisvad, J. C.; Hess, Philipp; Pouchus, Y. F.; Grovel, Olivier.
In order to assess the putative toxigenic risk associated with the presence of fungal strains in shellfish-farming areas, Penicillium strains were isolated from bivalve molluscs and from the surrounding environment, and the influence of the sample origin on the cytotoxicity of the extracts was evaluated. Extracts obtained from shellfish-derived Penicillia exhibited higher cytotoxicity than the others. Ten of these strains were grown on various media including a medium based on mussel extract (Mytilus edulis), mussel flesh-based medium (MES), to study the influence of the mussel flesh on the production of cytotoxic compounds. The MES host-derived medium was created substituting the yeast extract of YES medium by an aqueous extract of mussel tissues, with...

Determining sedimentary material sources in a Brazilian urban lake using Zn stable isotope compositions of bottom sediments: a preliminary study ArchiMer
Ferreira Araujo, Daniel; Boaventura, Geraldo; Mulholland, Daniel; Garnier, Jeremie; Machado, Wilson; Babinski, Marly; Dantas, Elton.
Zinc isotopes ratios provide qualitative and quantitative information about contributions from different Zn sources in the environment. Here we present the results of a preliminary study to identify the main sources controlling Zn isotope signatures of sediments from Lake Paranoá (Brazil), an urban water reservoir. The Zn isotope ratios were measured by MC-ICP-MS and expressed as δ66/64Zn notation relative to the JMC 3-0749-L solution. The studied sediments showed a narrow isotopic range (+0.00 to +0.13 ‰; n = 10) similar to that reported for intensely weathered soils worldwide (from -0.4 to +0.2‰). The geoaccumulation index (Igeo) also suggests low or negligible influence of Zn anthropogenic source. This research provides a baseline assessment of the Zn...

ABSTRACT Bloodstream infections (BSIs) are serious infections associated with high rates of morbidity and mortality. Every hour delay in initiation of an effective antibiotic increases mortality due to sepsis by 7%. Turnaround time (TAT) for conventional blood cultures takes 48 h, forcing physicians to streamline therapy by exposing patients to broad-spectrum antimicrobials. Our objective was (1) to evaluate the accuracy and TAT of an optimized workflow combining direct matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF MS) and in-house real-time polymerase chain reaction (PCR) for bacterial identification and antimicrobial resistance profiling directly from positive blood bottles for diagnosing bloodstream infections...
